2年後の研究結果:

・慢性移植片対宿主病(GvHD)の累積罹患率は、Grafalon(R) 非投与群の68.7%に対して、Grafalon(R) を投与した試験群は、32.2%でした。

・2年後の慢性GvHD無発症および無再発生存率の複合エンドポイントの率は、Grafalon(R) 非投与群(16.8%)よりもGrafalon(R) 投与群(36.6%)で有意に高くなりました。

・再発、感染合併症、急性GvHD、あるいは有害事象の率においては両群での有意差はありませんでした。

当研究の責任者を務めた、ドイツのハンブルグ大学病院(University Hospital Hamburg)幹細胞移植部門医長兼EBMT慢性悪性腫瘍作業部会(EBMT Chronic Malignancies Working Group)会長のNicolaus Kroeger教授は次のようにコメントしています。「多施設共同研究の結果では、再発の明らかなリスクを伴わずに慢性GvHDの発症リスクを低減することで、HLA適合同胞からの同種幹細胞移植を受けた白血病患者の転帰が大幅に改善しており、これにより移植後の患者の生活の質が向上し我々の日々の治療が変わってきます。」

当研究について

HLA適合同胞からの同種抹消血幹細胞移植の前処置レジメンの一部としてのGrafalon(R)の前向き、多施設共同、非盲検、無作為化第III相試験。当研究は、168名の登録患者を対象にして欧州の27施設で実施されました。患者は、無作為にGrafalon(R)の投与、非投与に割り当てられました。

GvHDについて

European Study in Patients With Acute Leukemia Shows the Inclusion of Grafalon(R) (Anti T-lymphocyte Immunoglobulin) in a Conditioning Regimen Before Stem Cell Transplantation From HLA-Identical Siblings Results in a Significantly Lower Incidence of Chronic GvHD

     Results of a European study, which was jointly funded by Neovii and the

European Society for Blood and Marrow Transplantation, have just been published

as a full manuscript in the January 2016 edition of the New England Journal of

Medicine.

    The results showed at 2 years:

    - The cumulative incidence of chronic GvHD was 32.2% in the study group

with Grafalon(R) compared to 68.7% in the non Grafalon(R) group.

     - The rate of the composite endpoint of chronic GvHD free and relapse free

survival at 2 years was significantly higher in the Grafalon(R) group than in

the group without (36.6% vs 16.8%).

     - There were no significant differences between the groups in rates of

relapse, infectious complications, acute GvHD, or adverse events.

    The principal investigator of the study, Professor Nicolaus Kroger, Medical

Director of the Department of Stem Cell Transplantation at the University

Hospital Hamburg, Germany and Chair of the EBMT Chronic Malignancies Working

Group commented "The results of our multicenter study significantly improved

the outcome of leukemia patients receiving an allogeneic stem cell transplant

from their HLA-identical sibling by reducing the risk of chronic GvHD without

obvious risk of relapse, which improves the quality of life of our patients

after the transplant and will change our daily practice."

    About the study

    Prospective, multicenter, open-label, randomized, phase 3 study of

Grafalon(R) as part of a conditioning regimen prior to transplantation of

allogeneic peripheral blood stem cells from HLA-identical siblings. The study

was conducted in 27 European centres and enrolled 168 patients. Patients were

randomized to either receive or not receive Grafalon (R)About GvHD

    Graft versus host disease (GvHD) is a serious, life threatening

complication after allogeneic stem cell transplantation. It develops, when the

new immune system, which arises from the transplanted stem cells (graft),

attacks tissues and organs of the recipient (host). It can be classified as

acute or chronic, depending on the time of occurrence and/or the pathology.
